Abstract FRET studies were performed on freely diffusing RNase H molecules at pH 7.2 and ph 3.0. The application of ALEX and PIE illumination and detection schemes allowed the accumulation of donor-only free smFRET data with excellent statistics. By imposing physically sensible global restrictions on the fit of the data, an intermediate folding state can be observed and characterized.
